Glycolipids as receptors for Bacillus thuringiensis crystal toxin.

چکیده
The development of pest resistance threatens the effectiveness of Bacillus thuringiensis (Bt) toxins used in transgenic and organic farming. Here, we demonstrate that (i) the major mechanism for Bt toxin resistance in Caenorhabditis elegans entails a loss of glycolipid carbohydrates; (ii) Bt toxin directly and specifically binds glycolipids; and (iii) this binding is carbohydrate-dependent and relevant for toxin action in vivo. These carbohydrates contain the arthroseries core conserved in insects and nematodes but lacking in vertebrates. We present evidence that insect glycolipids are also receptors for Bt toxin.

Investigation of Cytocidal Activity of Bacillus Thuringiensis Parasporal Toxin on CCRF-CEM Cell Line
Background & Objective: Parasporin is a parasporal protein of Bacillus thuringiensis and exhibits special cytocidal activity against human cancer cells. Similar to other insecticidal Bacillus thuringiensis crystal toxins, parasporin shows target specificity and damages the cellular membrane. In this study, different strains of Bacillus thuringiensis isolated from various regions of Iran and the...
متن کامل[21] Caenorhabditis elegans Carbohydrates in Bacterial Toxin Resistance
The major virulence factor produced by the bacterium Bacillus thuringiensis (Bt) is a pore‐forming toxin called crystal (Cry) toxin, which targets and kills insects and nematodes. To understand how this bacterial toxin interacts with its invertebrate hosts, a genetic screen in C. elegans for nematodes resistant to Bt toxin was carried out. Four of the five genes that mutated to toxin resistance...
متن کاملResistance to Bacillus thuringiensis toxin in Caenorhabditis elegans from loss of fucose.
A mutation in the Caenorhabditis elegans bre-1 gene was isolated in a screen for Bacillus thuringiensis toxin-resistant (bre) mutants to the Cry5B crystal toxin made by B. thuringiensis. bre-1 mutant animals are different from the four other cloned bre mutants in that their level of resistance is noticeably lower. bre-1 animals also display a significantly reduced brood size at 25 degrees C. He...
متن کاملRole of receptors in Bacillus thuringiensis crystal toxin activity.
Bacillus thuringiensis produces crystalline protein inclusions with insecticidal or nematocidal properties. These crystal (Cry) proteins determine a particular strain's toxicity profile. Transgenic crops expressing one or more recombinant Cry toxins have become agriculturally important. Individual Cry toxins are usually toxic to only a few species within an order, and receptors on midgut epithe...
متن کاملEffects of Protoplast Fusion on δ-endotoxin Production in Bacillus thuringiensis Spp. (H14)
In this study, mutant forms of Bacillus thuringiensis spp. israelensis (H14) were produced. These mutants were identified when the cells were cultured on chloramphenicol plates and stained with crystal violet. Protoplasts of the mutants were isolated by enzymatic digestion (lysozyme) of the cell walls at the presence of an osmotic stabilizer. The protoplasts were induced to fuse to each other i...
